The test uses histidine-deficient Salmonella Typhimurium strains or tryptophan-deficient Escherichia Coli strains. Because these mutant strains are unable to synthesize histidine or tryptophan on their own, they must rely on exogeneous histidine/tryptophan for growth as they cannot survive on selective media without histidine/tryptophan. When the mutagen is present, it causes a reverse mutation in the strain’s gene and transforms it from a nutrient-deficient strain to a wild-type that is able to grow and form visible colonies even on histidine/tryptophan deficient media.
