Human CD56+ NK cells
NK cells are derived from bone marrow lymphoid stem cells, whose differentiation and development depends on the bone marrow and thymus microenvironment. They are mainly distributed in the bone marrow, peripheral blood, liver, spleen, lungs, and lymph nodes. NK cells are different from T or B cells; they are a type of lymphocytes that can kill tumor cells and virus-infected cells non-specifically without sensitizing their target beforehand.
The targets of NK cells are mainly certain tumor cells (including some cell lines), virus-infected cells, certain self-tissue cells (e.g. blood cells), and parasites. As a result, NK cells are important factor in the body’s anti-tumor, anti-infection mechanism, and are also involved in type-II hypersensitivity reaction, graft-versus-host reaction.
In recent years, immune cell therapies have made rapid advancements in the treatment of diseases. For instance, scientists have genetically engineered T cells to recognize and attack tumor cells, a treatment known as CAR-T cell therapy, which has now become the mainstay of immunotherapy. NK cells are another important type of immune cells – a subpopulation of lymphocytes which are an essential part of the body’s intrinsic immunity and the first line of defense against viral infections and tumor cells. Based on this feature, CAR-NK cell therapy was also created. Compared with CAR-T cell therapy, CAR-NK cell therapy has fewer side effects and is inexpensive.
Supply of high-quality NK cells is crucial for CAR-NK cell therapy research. Human CD56+ NK cells produced by IPHASE are obtained from fresh human PBMC by immunomagnetic bead negative sorting. The cells are free of artifacts from selection process such as magnetic beads and antibodies. Furthermore, these cell products are ideal for subsequent downstream experiments such as cell culture or in-vitro drug metabolism due to its high purity.
