Peripheral blood mononuclear cell (PBMC) is a cell only one nucleus in the peripheral blood. Derived from hematopoietic stem cells (HSC) in the bone marrow, which usually includes mainly lymphocytes and monocytes, PBMC are distributed to tissues and organs throughout the body by blood circulation and are involved in immune response and immunomodulation.

    PBMC can mimic the in-vivo environment to provide an effective in-vitro model for drug development. Studying the effects of drugs on the targets and signaling pathways of PBMCs can assist in discovering new drug targets, and these discoveries can provide unprecedent ideas and directions for drug discovery and development.

    The main application directions of PBMC in immunotherapeutic drug development include:

    1.Antibody-dependent cell-mediated cytotoxicity (ADCC): PBMC plays a key role in ADCC; NK cells, macrophages, etc. are the main effector PBMCs in ADCC action. These cells can kill target cells encapsulated by antibodies through binding to them.

    2.Mixed Lymphocyte Reaction (MLR): a co-culture system for primary DC cells and T cells which can be used to evaluate DC-mediated T cell activation. DC cells express large quantities of PD-L1 in their surface, which can bind and inhibit PD-1 on the surface of T cells. If the drug can effectively block the reaction pathway between the two, then it can stimulate the proliferation of T cells and thus verify the functional activity of the drug.

    3.T cell activation assays: T cell activation assays are one of the commonly used assays for assessing the effects of immunotherapy drugs. In T cell activation assays, in-vitro cultured T cells are usually used and appropriate stimuli, such as antigens or antibodies, are given to induce the activation of T cells.

    4.T cell proliferation assay: T cell proliferation assay can be used for screening immunotherapy drug candidates and evaluating the effectiveness of the drugs by comparing the degree of T cell proliferation in different drug-treated experiment groups with the control group.

    Human PBMCs produced by IPHASE are isolated from human peripheral blood by density gradient centrifugation. These are single nucleus cells mainly composed of lymphocytes (T cells, B cells, and NK cells) and monocytes.
