    Glomerular mesangial cells are a kind of glomerular intrinsic cell located between the glomerular capillary loops and adjacent to endothelial cells or basement membrane. Due to their irregular morphology, cell protrusion can reach deep between endothelial cells and basement membrane, or extend into the capillary lumen between endothelial cells. The main functions of renal mesangial cells include maintaining the structural integrity of the glomerular capillary network, regulating the glomerular filtration rate, and secreting a variety of cell growth factors such as transforming growth factor β, platelet-derived growth factor, and biologically active substances such as renin, thereby performing a variety of biological functions. The afferent and efferent arterioles are connected to the glomerular mesangial cells at the entrance and exit of the glomerulus. Therefore, the contractile activity of the glomerular mesangial cells enhances the contraction of the arterioles, thus controlling the intraglomerular blood shunt.

    Normal human Kidney Mesangial cells (NhKMs) produced by IPHASE are obtained after cortical digestion and isolation of individual glomeruli. The glomeruli were further digested, isolated and purified through PDGF-Rβ positive selection to obtain NhKM cells, which were then cryopreserved to a total cell volume of 5×105/vial. The purity of cells was determined by immunofluorescence staining for PDGFRβ, vimentin and α smooth muscle actin. Normal human renal mesangial cells are terminally differentiated cells that can maintain a normal cell appearance even after 15 population doublings. NhKMs ​​have an intermediate phenotype with common characteristics of fibroblasts and smooth muscle cells.

    Quality test of normal human renal mesangial cells:

    Sterility: negative for mycoplasma, yeast and fungi test.

    Virus: negative for CMV, EBV HBV, HCV, HIV-1, HIV-2 test.

    Can be used for in-vitro studies of permeation and excretion, inflammation, and applied to drug screening/development such as hypertension, diabetes, oncology, autoimmune diseases, and toxicology screening.
