PART 01. In-Vitro Micronucleus Test
Micronuclei are whole chromatids or acentric fragments or ring chromosomes which remain in the cytoplasm when chromosomes regularly enter daughter cells to form nuclei after mitosis. Because these fragments or chromosomes cannot enter the main nucleus at telophase, when the daughter cells enter the next interphase, they condense into small nuclei outside the main nucleus and form micronuclei. With the elucidation of formation mechanism, the significance of micronuclei, the continuous improvement of detection method and perfection of experimental techniques, micronucleus test became one of the main methods for screening mutations. It is widely used for predicting carcinogenic risk, detecting aneuploidy inducers and other genetic hazards.
The in-vitro micronucleus test is a genotoxicity test method for detecting the production of micronuclei in mammalian cells after treated with a test substance. It is ideal for checking chromosome breakage and aneuploidy induced in mitotic cells during/after exposure to the test substance. Compared to the in-vivo test, the in-vitro micronucleus test is simpler, faster, and cheaper. In addition, it is not affected by factors such as individual differences between test animals. Since in-vitro micronucleus test can detect micronucleus reactions induced by certain chemicals at low concentrations with high sensitivity. Therefore, it has been widely used in the field of genetic toxicology.
PART 02. In vitro micronucleus test kit
The IPHASE in-vitro micronucleus test kit uses Chinese Hamster Lung (CHL) cells as test system. The CHL cells are exposed to the test substance with/without metabolic activation system and treated with actin polymerization inhibitor cytochalasin B. The cells are allowed to reach their ideal frequency of binucleated cells, and then harvested, prepared, and stained; then the micronucleus rate of cells which completed one mitosis (binucleated cells) is analyzed under microscope to evaluate the mutagenic potential of the test substance. IPHASE kit provides all essential reagents and cells for in-vitro micronucleus testing. Since all kits have passed strict quality testing, the produced experimental results are accurate, reliable and reproducible.
